    Changes over 26 Years in the Avifauna of the Bogotá Region, Colombia: Has Climate Change Become Important?

    High Neotropical mountains are among the most threatened ecosystems by climate change and this problem could be accentuated in cities where temperatures are higher. However, there are few data of long-term avifaunal changes in Neotropical cities, and the potential impact of climate change has yet to be addressed. Using data from 26 years of Audubon's Christmas Bird Counts (CBCs) in the Bogotá area (ca. 2,600–3,100 m in Colombia's eastern Andes), we analyze long-term changes of the avifauna using linear regressions corrected for species' habitat preferences and compared urban and rural sites. For the CBC we followed the National Audubon Society's methodology in which birds are counted over a 24-h period within a fixed circle 24 km in diameter. We recorded 235 species in the Bogotá circle with an average of 122 spp/year, including 46 boreal migrants, three endemic species, and four endemic subspecies, two globally and four locally threatened species. Species richness was higher in rural than in urban areas and most species were associated with native forest and scrub and wetlands. Among the species that were analyzed for changes in time 31% increased, 20% decreased, and 49% did not change. Strong fluctuations or changes in abundance were more frequent in urban than rural environments. Many of the species that increased or became established during the CBC interval came from lower elevations or the warmer, drier parts of the region beyond the count circle. By contrast, the lower elevational limits moved to higher elevations in several species that decreased. Climate change with its related effects represented the factor associated with the most changes in abundance. Other potential causes were direct human actions, mostly associated with urbanization, and “natural” successional changes in vegetation; predation by feral dogs and cowbird parasitism also affected several species negatively. Observations indicate that these factors will continue into the future, and the effects of climate change and urbanization in particular will probably intensify. The Bogotá CBC has provided much valuable information for Neotropical urban ornithology and its practice should be encouraged in other Latin American cities and should definitely continue in Bogotá for many years to come

    Distribution of \u3ci\u3eEscherichia coli\u3c/i\u3e Passaged through Processing Equipment during Ground Beef Production Using Inoculated Trimmings

    The contamination of raw ground beef by Escherichia coli O157:H7 is not only a public health issue but also an economic concern to meat processors. When E. coli O157:H7 is detected in a ground beef sample, the product lots made immediately before and after the lot represented by the positive sample are discarded or diverted to lethality treatment. However, there is little data to base decisions on how much product must be diverted. Therefore, five 2,000-lb (907-kg) combo bins of beef trimmings were processed into 10-lb (4.54-kg) chubs of raw ground beef, wherein the second combo of meat was contaminated with a green fluorescent protein (GFP)–expressing strain of E. coli. This was performed at two different commercial ground beef processing facilities, and at a third establishment where ground beef chubs from the second grinding establishment were mechanically split and repackaged into 3-lb (1.36-kg) loaves in trays. The GFP E. coli was tracked through the production of 10-lb (4.54-kg) chubs and the strain could not be detected after 26.5% more material (500 lb or 227 kg) and 87.8% more material (1,840 lb or 835 kg) followed the contaminated combo at each establishment, respectively. Three-pound (1.36-kg) loaves were no longer positive after just 8.6% more initially noncontaminated material (72 lb or 33 kg) was processed. The GFP strain could not be detected postprocessing in any residual meat or fat collected from the equipment used in the three trials. These results indicate that diversion to a safe end point (lethality or rendering) of the positive lot of ground beef, plus the lot before and lot after should remove contaminated ground beef, and as such provides support for the current industry practice. Further, the distribution and flow of E. coli on beef trimmings through various commercial equipment was different; thus, each establishment needs to consider this data when segregating lots of ground beef and establishing sampling protocols to monitor production

    Cambios en la avifauna de un relicto de bosque en la franja periurbana de Bogotá a lo largo de catorce años

    A lo largo de varios siglos, la vegetación nativa de la parte plana de la Sabana de Bogotá ha sido casi completamente sustituida por cultivos, potreros y urbanizaciones. El último relicto de esta vegetación es un pequeño bosque (ca 10 ha), ubicado en la Hacienda Las Mercedes en el borde norte de la ciudad de Bogotá. El tamaño reducido y aislamiento del bosque, agravado por el crecimiento descontrolado de vegetación invasiva (lianas y zarzamora silvestre) ha generado la pérdida de muchas especies. Sin embargo, en los últimos años el bosque ha sido objeto de acciones de rehabilitación y actualmente la zona está inmersa en una reserva en donde se prevén programas de restauración más amplios. Con el objetivo de evaluar los cambios en la comunidad de aves para estimar los efectos de las acciones de restauración, se registró la avifauna presente en 2001-2002 y en el 2014, mediante registros visuales y auditivos en puntos fijos en el bosque. En el primer censo se encontraron 27 especies de bosque y en el segundo 30, y las abundancias relativas de por lo menos la tercera parte de ellas también aumentó en los 13 años, indicando un resultado positivo en la recuperación del bosque. Los resultados resaltan la capacidad de recuperación de los ecosistemas degradados y la importancia de continuar con las acciones de restauración en el área de reserva.The native vegetation of the Sabana de Bogota’s flat area has been replaced by agricultural lands, pastures and urbanizations over the years. The last relict of this native vegetation is a small forest (ca 10 ha) at Hacienda Las Mercedes, in the northern border of Bogota. The forest’s small size and isolation from other areas of natural vegetation have resulted in the loss of many species. The problem was exacerbated by uncontrolled growth of invasive vegetation (vines and wild blackberry). However, in recent years restorative action including control of these vegetation has been undertaken and the forest is now immerse in a natural reserve where more restorative actions are being planned. Using point count stations the authors recorded all the birds seen and heard in 2001-2002 and in 2014 in order to evaluate the changes in the bird community and estimate the effects of the restorative actions. We recorded 27 forest species in the first census and 30 in the second. The relative abundance of at least 1/3 of the species also increased indicating a positive result in the forest restoration. The results highlight the capacity of degraded ecosystems to recover and the importance of continuing the restorative actions in the reserve area

    Distinct dynamics of social motivation drive differential social behavior in laboratory rat and mouse strains

    Mice and rats are widely used to explore mechanisms of mammalian social behavior in health and disease, raising the question whether they actually differ in their social behavior. Here we address this question by directly comparing social investigation behavior between two mouse and rat strains used most frequently for behavioral studies and as models of neuropathological conditions: C57BL/6 J mice and Sprague Dawley (SD) rats. Employing novel experimental systems for behavioral analysis of both subjects and stimuli during the social preference test, we reveal marked differences in behavioral dynamics between the strains, suggesting stronger and faster induction of social motivation in SD rats. These different behavioral patterns, which correlate with distinctive c-Fos expression in social motivation-related brain areas, are modified by competition with non-social rewarding stimuli, in a strain-specific manner. Thus, these two strains differ in their social behavior, which should be taken into consideration when selecting an appropriate model organism

    A Dynamic Programming Approach to Adaptive Fractionation

    We conduct a theoretical study of various solution methods for the adaptive fractionation problem. The two messages of this paper are: (i) dynamic programming (DP) is a useful framework for adaptive radiation therapy, particularly adaptive fractionation, because it allows us to assess how close to optimal different methods are, and (ii) heuristic methods proposed in this paper are near-optimal, and therefore, can be used to evaluate the best possible benefit of using an adaptive fraction size. The essence of adaptive fractionation is to increase the fraction size when the tumor and organ-at-risk (OAR) are far apart (a "favorable" anatomy) and to decrease the fraction size when they are close together. Given that a fixed prescribed dose must be delivered to the tumor over the course of the treatment, such an approach results in a lower cumulative dose to the OAR when compared to that resulting from standard fractionation. We first establish a benchmark by using the DP algorithm to solve the problem exactly. In this case, we characterize the structure of an optimal policy, which provides guidance for our choice of heuristics. We develop two intuitive, numerically near-optimal heuristic policies, which could be used for more complex, high-dimensional problems. Furthermore, one of the heuristics requires only a statistic of the motion probability distribution, making it a reasonable method for use in a realistic setting. Numerically, we find that the amount of decrease in dose to the OAR can vary significantly (5 - 85%) depending on the amount of motion in the anatomy, the number of fractions, and the range of fraction sizes allowed. In general, the decrease in dose to the OAR is more pronounced when: (i) we have a high probability of large tumor-OAR distances, (ii) we use many fractions (as in a hyper-fractionated setting), and (iii) we allow large daily fraction size deviations.Comment: 17 pages, 4 figures, 1 tabl

    Detection and monitoring of the multiple inflammatory responses by photoacoustic molecular imaging using selectively targeted gold nanorods

    In vitro cell experiments have been performed to detect and monitor the upregulation of intercellular adhesion molecule-1 (ICAM-1) and E-selectin simultaneously by photoacoustic molecular imaging (PMI). Human umbilical vein endothelial cells (HUVECs) were grown on gelatin-coated glass slides and stimulated with inflammatory cytokines to induce the expression of the inflammatory biomarkers, ICAM-1 and E-selectin. Gold nanorods (GNRs) of aspect ratio (AR) 1:3 with absorption centered at 715 nm conjugated to anti-ICAM-1 antibody and GNRs of AR 1:3.5 with absorption centered at 800 nm conjugated to anti-E-selectin were exposed to HUVECs with different stimulation conditions. A focused high frequency ultrasonic transducer (60 MHz, f/1.5) was used to scan the photoacoustic (PA) signal over the top surface of the cell containing slides. Averaged PA signal intensity from the stimulated cells was about 3 folds higher (~10 dB) compared to the un-stimulated cells for both ICAM-1 and E-selectin. The strong binding of GNRs to the stimulated HUVEC cells was evidenced by fluorescence imaging. Exposure of HUVEC cells to GNRs conjugated to isotype control antibodies confirms a low level non-specific binding. Also, at 0, 2, 6, and 24 hours after inflammatory stimulation, the HUVECs were exposed to GNRs conjugated anti-ICAM-1 antibody and anti-E-selectin antibody.     Tocilizumab in Hospitalized Patients with Severe Covid-19 Pneumonia

    BACKGROUND Coronavirus disease 2019 (Covid-19) is associated with immune dysregulation and hyperinflammation, including elevated interleukin-6 levels. The use of tocilizu- mab, a monoclonal antibody against the interleukin-6 receptor, has resulted in better outcomes in patients with severe Covid-19 pneumonia in case reports and retrospective observational cohort studies. Data are needed from randomized, placebo-controlled trials. METHODS In this phase 3 trial, we randomly assigned patients who were hospitalized with severe Covid-19 pneumonia in a 2:1 ratio receive a single intravenous infusion of tocilizumab (at a dose of 8 mg per kilogram of body weight) or placebo. Approxi- mately one quarter of the participants received a second dose of tocilizumab or placebo 8 to 24 hours after the first dose. The primary outcome was clinical status at day 28 on an ordinal scale ranging from 1 (discharged or ready for discharge) to 7 (death) in the modified intention-to-treat population, which included all the patients who had received at least one dose of tocilizumab or placebo. RESULTS Of the 452 patients who underwent randomization, 438 (294 in the tocilizumab group and 144 in the placebo group) were included in the primary and secondary analyses. The median value for clinical status on the ordinal scale at day 28 was 1.0 (95% confidence interval [CI], 1.0 to 1.0) in the tocilizumab group and 2.0 (non-ICU hospitalization without supplemental oxygen) (95% CI, 1.0 to 4.0) in the placebo group (between-group difference, −1.0; 95% CI, −2.5 to 0; P=0.31 by the van Elteren test). In the safety population, serious adverse events occurred in 103 of 295 patients (34.9%) in the tocilizumab group and in 55 of 143 patients (38.5%) in the placebo group. Mortality at day 28 was 19.7% in the tocilizumab group and 19.4% in the placebo group (weighted difference, 0.3 percentage points (95% CI, –7.6 to 8.2; nominal P=0.94). CONCLUSIONS In this randomized trial involving hospitalized patients with severe Covid-19 pneu- monia, the use of tocilizumab did not result in significantly better clinical status or lower mortality than placebo at 28 days.
